Effects of Tamsulosin, an Α1-adrenergic Antagonist, and Tak- 802, a Novel Acetylcholinesterase Inhibitor, and Their Synergistic Effects on the Urodynamic Characteristics in a Guinea Pig Model of Functional Bladder Outlet Obstruction

چکیده

Hypothesis / aims of study The efficacy of α1-adrenergic antagonists in the treatment of lower urinary tract symptoms (LUTS) in patients with benign prostatic hyperplasia (BPH) is generally attributable to improvement of the urodynamic characteristics and relief of bladder instability. While the latter effect has been partly confirmed in an animal model with partial bladder outlet obstruction (BOO), the former effect has not been well documented in experimental studies using animals, because no suitable animal model has yet been developed to assess the effects of α1-adrenergic antagonists on the urodynamic characteristics. In this study, we attempted to establish an animal model with functional BOO to model the dynamic component of BPH, and to evaluate the effects of tamsulosin, an α1-adrenergic antagonist, TAK-802, a novel acetylcholinesterase (AChE) inhibitor with some selectivity for muscarinic actions (1), and of both administered concomitantly on the urodynamic characteristics in this model.

In this study the effects of bromocriptine and apomorphine (dopamine receptor agonists) on electrical field induced twitch response of longitudinal muscle of guinea-pig illeum was investigated. Bromocriptine and apomorphine dose dependently inhibited illeal contraction. IC50 for this inhibitory effects were 6.22±0.645×10^-7 M and 5.48±0.647×10^-6 M, respectively. sulpiride (a specific D2 dopami...

Effects of Tamsulosin on the Urinary Bladder Function in Rats with Partial Outlet Obstruction

Hypothesis / aims of study Overactive bladder (OAB) associated with bladder outlet obstruction (BOO) is common in elderly men. However, its exact etiology remains unclear. Tamsulosin is an α1-adrenoceptor blocker used in the treatment of symptomatic benign prostatic hyperplasia worldwide, and known to be effective for OAB as well as obstructive symptoms. In this study, we investigated the effec...

BRONCHODILATORY AND AN TICHOLINERGIC EFFECTS OF Carum copticum ON ISOLATED GUINEA PIG TRACHEAL CHAINS

Carum copticum is a grassy and annual plant which is believed to have several therapeutic effects, including anti-asthmatic properties. We have therefore studied the bronchodilatory and anticholinergic effects of 4 rnL aqueous extract (AE), 0.0 5 mL ethanol extract (EE), and 0.0 0 5 mL essential oil (EO) of Carum copticum and 1 rnL of a 5 mM solution of its main ingredient, thymol (T), in ...

Effects of alpha 1-adrenergic receptor subtype selective antagonists on lower urinary tract function in rats with bladder outlet obstruction.

PURPOSE Antagonists of alpha 1-adrenergic receptors (alpha 1ARs) relieve obstructive and irritative symptoms in patients with bladder outlet obstruction. However, to our knowledge mechanisms underlying the relief of irritative symptoms remain unknown. Because bladder alpha 1dARs are up-regulated in some rats with bladder outlet obstruction, we investigated the effect of the alpha 1aAR antagonis...

Effects of Spinal and Peripheral Injection of α1A or α1D Adrenoceptor Antagonists on Bladder Activity in Rat Models with or without Bladder Outlet Obstruction

PURPOSE Antagonists of α1-adrenergic receptors (α1ARs) relax prostate smooth muscle and relieve voiding and storage symptoms. Recently, increased expression of α1ARs with change of its subtype expression has been proved in bladder outlet obstruction (BOO). To search for the evidence of changes in α1ARs subtype expression and activity in the peripheral and spinal routes, the effects of spinal an...
